مقایسه اثر عصاره آبی ریشه ارکید (Dactylorhiza maculate) با ایبوبروفن بر آستانه حسی درد با استفاده از آزمایش فرمالین در موش صحرایی نر بالغ

چکیده

مقدمه: درد احساس نامطبوعی است که معمولاً در اثر صدمات وارده به بافت ها به وجود می ­آید. گیاه ارکید با داشتن گلوکومانان دارای اثرات ضددردی در مدل های ایجاد درد حاد و مزمن است. با توجه به خواص آنتی اکسیدانی و ضدالتهابی ترکیبات موجود در ریشه این گیاه، هدف از انجام این طرح بررسی اثر عصاره ریشه ارکید بر کاهش و پیشگیری از درد فرمالینی بود.
روش کار: در این مطالعه تجربی 56 سر موش صحرایی نر نژاد ویستار به طور تصادفی در 7 گروه کنترل، شاهد، ایبوبروفن (6 میلی گرم بر کیلوگرم) و چهار گروه دریافت کننده عصاره آبی ریشه ارکید (80، 160، 320 و 640 میلی گرم بر کیلوگرم) قرار گرفتند. تزریق ها به صورت درون صفاقی انجام شد. سی دقیقه پس از تزریق عصاره ارکید، آزمایش فرمالین انجام شد. داده ها با استفاده از آزمون آماری تحلیل واریانس یک طرفه و آزمون تعقیبی دانکن در سطح معناداری 0.05 تجزیه و تحلیل شدند.
یافته­ ها: غلظت­ های 320 و 640 میلی ­گرم بر کیلوگرم باعث کاهش معنادار درد حاد و غلظت­ های 160، 320 و 640 میلی­ گرم بر کیلوگرم باعث کاهش معنادار درد مزمن شد. ایبوبروفن نیز درد حاد و مزمن ناشی از فرمالین را کاهش ­داد.
 نتیجه گیری: نتایج این مطالعه نشان داد که عصاره ریشه ارکید به صورت وابسته به دوز دارای خاصیت ضد دردی است که احتمالا به خاصیت آنتی­ اکسیدانی و خاصیت ضد التهابی آن و همچنین وجود گلوکومانان مربوط می شود.

چکیده [English]

Introduction: Pain is an unpleasant feeling that usually occurs as a result of tissue injuries. Orchid could have significant analgesic effects in acute and chronic pain modalities due to its glucomannan content. Considering the antioxidant and anti-inflammatory properties of the Orchid root’s compounds, the aim of this study was to determine the effect of Orchid root extract on the reduction and prevention of formalin-induced pain.
Materials and Methods: In this experimental study, 56 male Wistar rats were randomly divided into 7 groups, including Control, Sham (distilled water), Ibuprofen (6 mg/kg) groups and four groups receiving the aqueous extract of Orchid root (80, 160, 320 and 640 mg/kg). All treatments were injected intra peritoneal. Formalin test was performed 30 minutes after injection of Orchid extract. Data were analyzed by one-way ANOVA and Duncan's post-test at a significance level of 0.05.
Results: Orchid at concentrations of 320 and 640 mg/kg caused a significant reduction in acute pain and at concentrations of 160, 320 and 640 mg/kg significantly decreased chronic pain. Ibuprofen also reduced acute and chronic pain induced by formalin.
Conclusion: This research demonstrated the dose-dependent anti-nociceptive effect of Orchid root extract which is
probably related to glucomannan and its antioxidant and anti-inflammatory properties.
