اثر عصاره سرخه‌ولیک (Crataegus monogyna) بر فاکتورها و ساختار هیستولوژیکی کلیه در موش‌های صحرایی دیابتی

چکیده
مقدمه: عوارض کلیوی ناشی از دیابت از علل مهم مرگ‌ و‌ میر افراد دیابتی است. در مطالعه حاضر اثر عصاره هیدروالکلی سرخه‌ولیک بر فاکتورهای کلیوی و ساختار هیستولوژیکی کلیه در موش‌های صحرایی دیابتی بررسی شد.
روش کار: در مطالعه تجربی حاضر 40 موش‌ صحرایی نر نژاد ویستار به طور تصادفی در5 گروه 8 تایی شامل گروه های کنترل (دریافت نرمال سالین)، دیابتی (دریافت استرپتوزوتوسین(STZ))، تیمار (دریافت STZ و عصاره سرخه‌ولیک در دوزهای 100، 200 و 400 mg/kg) قرار گرفتند. تیمار توسط عصاره و دارو، 72 ساعت بعد از تزریق داخل صفاقی تک دوز STZ (60mg/kg) به مدت سه هفته انجام شد. گلوکز، اوره و کراتینین سرم اندازه ­گیری شد و سپس بافت کلیه تحت بی‌هوشی عمیق جدا و برای مطالعات هیستوپاتولوژی استفاده شد.
یافته ها: میزان گلوکز در روز 21 پس از تیمار با عصاره سرخه ­ولیک، کاهش و میزان اوره در تمام گروه ­های تیمار با عصاره نسبت به گروه دیابتی افزایش یافت. دوز 100 و 400 (mg/kg) عصاره سرخه­ ولیک، سبب کاهش معنا­داری در میزان کراتینین نسبت به گروه دیابتی شد. در گروه دیابتی تخریب کامل بافت کلیه در کورتکس و تخریب کامل و نکروز صد درصدی نفرون‌ها مشاهده شد. بافت کلیه در موش‌های دیابتی تحت تیمار توسط عصاره طبیعی بود و پرخونی و نکروز نفرون‌ها مشاهده نشد و جسمک‌های کلیوی طبیعی بودند.
نتیجه گیری: عصاره سرخه‌ولیک می ­تواند سبب کاهش کراتینین گردد درحالی که میزان اوره را افزایش می ­دهد که ممکن است ناشی از مقدار عصاره به­ کار رفته باشد. هم ­چنین می ­تواند بر پیشگیری از تخریب بافتی کلیه در اثر دیابت موثر باشد.

چکیده English

Introduction: Renal complications is considered as important cause of mortality in diabetic patients. This study
investigated the effect of hydroalcoholic extract of C. monogyna on renal histopathological changes in diabetic rats.
Materials and Methods: In this experimental study 40 male Wistar rats were divided into 5 groups (n= 8) including control (receiving normal saline), diabetic (receiving streptozotocin), treatment groups (receiving STZ and Crataegus monogyna (C. monogyna) extract at 100, 200 and 400 mg/kg). Treatments with drug and extract were performed 72 hours after intra-peritoneal injection of a single-dose of STZ (60 mg/kg). Glucose, urea and ceratinine in serum were meseared and then kidney tissue was removed under deep anesthesia and used for histopathological studies.
Results: The amount of glucose increased on day 21 after treatment with extract C. monogyna and the level of urea in all treatment groups with extract increased compared to diabetic group. The dose of 100 and 400 mg / kg of extract reduced the creatinine level significantly compared to the diabetic group. In the diabetic group, complete destruction of the renal tissue in cortex and complete degeneration and 100% necrosis of the nephrons were observed. In diabetic rats treated with C. monogyna extract, kidney tissue was normal and no renal bleeding and necrosis were observed, and kidney cysts were normal.
Conclusion: C. monogyna extract was effective in preventing renal tissue damage due to diabetes.
