اثرات ضد دردی کلرفنیرامین در بیهوشی موش صحرائی با ترکیب آسپرومازین- کتامین

چکیده

زمینه و هدف: اخیراً آنتی هیستامین ها به عنوان عوامل کاهش دهنده درد مورد توجه قرار گرفته اند. هدف از مطالعه حاضر، تحقیق  ایجاد  بی دردی کلرفنیرامین در طی بیهوشی با ترکیب آسپرومازین- کتامین و انجام عمل جراحی در موش صحرائی بود.

مواد و روش کار: تعداد 20 سر موش صحرائی بالغ به صورت تصادفی به دو گروه تقسیم شدند.   درگروه اول کلرفنیرامین تزریق شده و 30 دقیقه بعد آسپرومازین - کتامین تزریق شد؛ در گروه دوم  ابتدا نرمال سالین تزریق شد و سپس ترکیب آسپرومازین - کتامین تزریق گردید. ارزیابی رفلکس های درد از طریق آزمون فشردن پنجه پا ، دم و واکنش موش صحرائی به عمل لاپاراتومی در طی بیهوشی صورت گرفت. زمان القاء بیهوشی، طول دوره بیهوشی سبک، زمان بیهوشی جراحی، مدت زمان لازم برای راه رفتن، تعداد ضربان قلب و تنفس اندازه گیری شد.  

نتایج: طول مدت بیهوشی سبک در گروه 2 کوتاهتر از گروه 1  بود (05/0 P <). رتبه درد لاپاراتومی در گروه اول و گروه دوم از نظر آماری تفاوت معنی دار با همدیگر داشتند. در دو گروه  رفلکس درد ناشی از فشردن پنجه پا و دم تفاوت معنی دار با همدیگر نداشتند. ضربان قلب در موش های صحرائی در هر دو گروه در مقایسه با هم تغییر آماری معنی دار نشان نداد.

نتیجه گیری: کلرفنیرامین به عنوان یک داروی پیش بیهوشی با ترکیب آسپرومازین- کتامین در موش صحرائی بیهوشی متعادل مناسبی را القاء می کند.  به این ترتیب، کلرفنیرامین در موش صحرائی می تواند به عنوان یک ماده پیش بیهوشی در ترکیب با آسپرومازین- کتامین در اعمال جراحی استفاده شود.

Background & Objective: Recently, antihistamines is known as analgesic agents. The aim of the present study is to investigate the  analgesic activity of chlorpheniramine (H1 antagonist), during anesthesia with acepromazine- ketamine combination  and surgical operation in rat.

Methods & materials: 20 adult rats were randomly assigned in two groups. In  first group,  chlorpheniramine was administered  and 30 minutes later acepromazine – Ketamine was  injected.   In second  group, Normal saline was administerd  initially  and  then acepromazine - Ketamine combination was injected.   Assessment of pain reflexes was performed  by toe pinch , tail pinch test and reflex of rats to laparotomy operation during anesthesia. Induction time of anesthesia, duration of light anesthesia, surgical anesthesia time, walking time, heart rate and respiratory rate was measured. 

Results: Time of light anesthesia in group 2    was shorter than group 1 (P < 0.05). Laparotomy pain  score was significantly different in first group and second group. Toe pinch and tail reflex didn’t show significant differences between two groups. Heart rate in all rats not showed significant differences in compared together .  

Conclusion: chlorpheniramine as a premedication was induced  appropriate balance anesthesia in rat with acepromazine- ketamine combination. Thus, chlorpheniramine could be used in rat as premedication agent for surgical operations in combined with acepromazine- ketamine.
