اثر عصار آبی گیا مفری بر کولیت زخمی در موشها ی صحرایی ونر بالغ

چکیده
مقدمه: کولیت زخمی (Ulcerative colitis) یک بیماری مزمن التهابی است که مخاط و زیر مخاط را در کولون و رکتوم تحت تاثیر قرار میدهد. اتیوپاتولوژی این بیماری را به عدم تعادل پاسخ­های ایمنی مخاط و فلور باکتریایی مقیم مخاط و همچنین فاکتور­های محیطی و ژنتیک مرتبط می دانند .کولیت زخمی با علائمی از جمله: دردهای شکمی؛اسهال و خونریزی های مقعدی مشخص میشود. از طرفی گیاه مفروDracocephalum polychaetum  به عنوان دارو به طور سنتی جهت بیماری­های گوارشی استفاده میشده است؛ لذا هدف مطالعه حاضر برای اثر عصاره آبی گیاه مفرو بر کولیت القاء شده بوسیله اسید استیک میباشد.

روش کار: در این مطالعه تجربی 48 عدد از موش­های صحرایی نر بالغ نژاد Wistar  درمحدوده وزنی280-230 گرم مورد آزمایش قرار گرفتند. موش ها پس از القای کولیت به گروههای، دوزهای متفاوت عصاره mg/kg)50،100،200(، شاهد، داروی پردنیزولون و آخرین گروه به عنوان گروه کنترل  تقسیم شدند. برای القاء کولیت، موشهای صحرایی مدت 36 ساعت از غذا محروم شدند و سپس تحت یک بی هوشی خفیف باCO2 ، میزان 2 میلی لیتر اسید استیک 4 درصد از طریق مقعد وارد کولون گردید. 24 ساعت بعد مشاهدات ماکروسکوپی مدفوع علائم کولیت را نشان داد. ارزیابی بیوشیمیایی از کولون ملتهب با سنجش فعالیت میلوپراکسیداز و میزان پراکسیداسیون لیپیدها صورت گرفت.

یافته ها: عصاره گیاه مفرو در دوز های (mg/kg 50 (001/0>P < /span>)200؛(001/0>P < /span>)100؛(01/0>P < /span>)) و پردنیزولون ( 001/0>P < /span> ) کاهش  نسبت وزن به طول کولون  در مقابل گروه شاهد را نشان می دهند.  عصاره با دوز mg/kg) 200) (05/0>P < /span>) و گروه پردنیزولون (001/0>P < /span>) شدت و میزان  التهاب را در مقایسه با گروه شاهدکاهش دادند.  غلظت MDA در گروه بیمار با اسید استیک افزایش یافت در حالیکه کاهش معناداری در حیوانات تحت درمان  با عصاره D.Polychaetum و پردنیزولون در سطح (01/0>P < /span>) نشان داد.

بحث و نتیجه­گیری: می توان نتیجه گرفت که عصاره گیاه مفرو بر درمان کولیت القاء شده بوسیله اسید استیک  موثر میباشد.میتوان گفت عصاره گیاه مفرو خاصیت ضد التهابی دارد.

Effect of Dracocephalum polychaetum Burnm, aqueous extract on acetic acid-induced ulcerative colitis in adult male rats

چکیده English

     Background: Ulcerative colitis (UC) is an inflammatory chronic disease that affects mucosa and submucosa  of the colon and rectum. The etiopathology is related to imbalance of the mucosal immune respanse toward the resident bactorical flora together with genetic and envirmental factors. UC is thogather with annoying symptoms such as; abdominal pain, diarrhea, and rectal bleeding. In other hand plant of Dracocephalum polychaetum have used traditionally in treat of digestive tract disease and as tranquillizing. Therefore the aim of the present study, was to evaluated the effect of this herbal aqueous extract on a model of acute experimental colitis.

      Methods: This experimental study were performed on 48 mail wistar rats(230-280g).Then induced ulcerative colitis, rats divided to groups incloude doses of extract (50, 100,200 mg/kg), vehicle,prednisolon and the last considered as control group.For induction of colitis the rats were fasted for 36 hours and then anaesthetized with CO2,at the last stage 2 ml of acetic acid 4% was instilled via the anus. After 24 hours the macroscopic study of colon showed the colitis indices. Biochemical evalution of inflamed colon was done using assay of myeloperoxidase (MPO) activity and  rate of lipid peroxidation.

    Results: The aqueous extract of D.polychaetum with doses of (50(P < span dir="RTL">>0/01),100(P < span dir="RTL">>0/001),200(P < span dir="RTL">>0/001)mg/kg) significantly reduced colon weight/length ratio. Extract at dose of 200mg/kg (P < span dir="RTL">>0/05) was effective to decrease inflammation as well as extent of injury. Concentration of malondialdehyde (MDA) were increased in acetic acid-treated groups‚ while recovered by treatment of animals with D.polychaetum (200 mg/kg) (P < span dir="RTL">>0/01) as well as prednizolon (10 mg/kg).

    Conclusion: It is concluded that Dracocephalum polychaetum Burnm aqueous extract could be effective in treatment against experimental acute colitis. The results suggested extract had anti-inflammatory effect.
