تاثیر پرتودهی سلول های سرطانی رده MCF-7 بر فعال شدن مسیر گیرنده شبه NOD : رویکرد بیوانفورماتیکی

چکیده
مقدمه: سرطان سینه از شایع‌ترین سرطان‌ها در زنان جهان است. مطالعه حاضر داده های ریزآرایه سلول‌های MCF-7 که به مدت 72 و 96 ساعت پرتودهی شده اند انالیز مجدد نمود تا مسیرهای سلولی مهم تشخیص داده شوند.
روش کار: داده‌های ریز آرایه سلول های MCF-7 که در دو بازه زمانی 72 و 96 ساعت تحت پرتو یونیزان با شدت 6Gy قرار گرفته بودند انتخاب و توسط نرم افزارR مورد تحلیل قرار گرفتند. سپس ژن‌هایی که تفاوت بیان داشتند استخراج و تحلیل بیان ژن در مورد آن‌ها انجام شد.
یافته‌ها: در این مطالعه پس از 72 و 96 ساعت پرتودهی به ترتیب 274 و 531 ژن با Log2FC>1 افزایش بیان و 174 و 649 ژن با Log2FC<-1 کاهش بیان داشتند (Adjusted p value <0.05 ). آنالیز عملکرد نشان داد که پس از 72 ساعت پرتودهی ژن های افزایش بیان یافته با مسیر پیام رسانی گیرنده شبه NOD مرتبط می باشد. ژن های استخراج شده در این مسیر شامل TXNIP ، IRF3/7، OAS ، TNFα ، LC3، GBPs ، CXCL،IL8 ، IRF9،JAK1 و STAT1/2 بودند (Adjusted p value <0.05 ). همچنین یافته ها نشان می دهد که پس از 96 ساعت پرتودهی بیشترین تعداد فرآیند بیولوژیک مشاهده شده برای ژن هایی که افزایش بیان داشته اند در ارتباط با انتقال سیگنال می باشد..
نتیجه‌گیری: تحلیل ژن‌های افزایش بیان یافته پس از ۷۲ ساعت پرتودهی، فعال شدن مسیر پیام رسانی گیرنده شبه NOD را نشان داد که احتمالا می تواند مرتبط با مسیرهای مولکولی آسیب به DNA و فعال‌سازی پاسخ‌های ایمنی ذاتی باشد.

Introduction:
Breast cancer is one of the most common cancers among women worldwide. In the present study, microarray data from MCF-7 cells irradiated for 72 and 96 hours were reanalyzed to identify important cellular pathways.
Methods:
Microarray data of MCF-7 cells that were exposed to ionizing radiation at an intensity of 6Gy for two time periods of 72 and 96 hours were selected and analyzed using R software. Then, genes that had differential expression were extracted and annotation analysis was performed on them.
Results:
In this study, after 72 and 96 hours of irradiation, 274 and 531 genes were upregulated with Log2FC>1 and 174 and 649 genes were downregulated with Log2FC<-1, respectively (Adjusted p value <0.05). Functional analysis showed that after 72 hours of irradiation, the upregulated genes were related to the NOD-like receptor signaling pathway. The genes extracted in this pathway included TXNIP, IRF3/7, OAS, TNFα, LC3, GBPs, CXCL, IL8, IRF9, JAK1 and STAT1/2 (Adjusted p value <0.05).The findings also show that after 96 hours of irradiation, the largest number of biological processes observed for upregulated genes were related to signal transduction.
Conclusion:
Pathway analysis on up-regulated genes after 72 hours of irradiation showed activation of the NOD-like receptor signaling pathway, which could possibly be related to molecular pathways of DNA damage and activation of innate immune responses.
