بررسی تغییرات بیان ژنی فاکتور رشد تبدیل کننده β1 و سیرتوئین1 در بافت کلیه پس از 8 هفته تمرین تناوبی شدید و فیستین در موش های مبتلا به دیابت نوع 2

چکیده
مقدمه: فاکتور رشد تبدیل کننده β1 مولکولی کلیدی در فیبروز کلیوی ناشی از دیابت است و درک مکانیسم‌های مختلف مداخله‌های حفاظت‌کننده بافت کلیه اهمیت بسزایی دارد. در این مطالعه تغییرات بیان ژنی فاکتور رشد تبدیل کنندهβ1 و سیرتوئین1 بافت کلیه متعاقب تمرین تناوبی شدید و فیستین در موش‌های دیابتی بررسی شد.
روش کار: 30 سر موش نر گونه C57bl/6 به گروه‌های اصلی کنترل با رژیم غذایی نرمال(6 سر) و پرچرب (24 سر) تقسیم شدند. دیابت نوع2 با 16 هفته مصرف رژیم غذایی پرچرب القا شد. سپس موش‌های دیابتی بطور تصادفی به گروه‌های دیابت، دیابت-تمرین، دیابت-فیستین، دیابت-تمرین-فیستین تقسیم شدند. 20 میلی‌گرم/کیلوگرم فیستین روزانه، طی8 هفته تزریق درون‌صفاقی شد. پروتکل تمرینی، شامل 8 هفته دویدن با وهله‌های 2 دقیقه‌ای با شدت 80-110% توان هوازی حداکثر با وهله‌های استراحت دو دقیقه‌ای در شدت50 % توان هوازی حداکثر و 5 روز/هفته بود. آنالیز داده‌ها با استفاده از آزمون تحلیل واریانس یک‌راهه انجام شد(0.05>P).
یافته ها: با القای دیابت بیان ژنی فاکتور رشد تبدیل کنندهβ1 در بافت کلیه افزایش و سیرتوئین1 کاهش یافت(001/0>P). هم‌چنین تمرین تناوبی شدید، فیستین و مداخله ترکیبی منجر به تغییر معنادار بیان ژنی فاکتور رشد تبدیل کنندهβ1 و سیرتوئین1 شد اما مداخله ترکیبی با تاثیر معناداری قوی‌تری در مقایسه با دو مداخله دیگر شد (0.001>P).
نتیجه‌گیری: به نظر می‌رسد تمرین تناوبی شدید و فیستین ، می‌توانند با افزایش سیرتوئین1 منجر به کاهش آسیب کلیوی ناشی از دیابت نوع 2، حداقل بواسطه کاهش بیان ژنی فاکتور رشد تبدیل کنندهβ1 شوند. هر چند مداخله ترکیبی با تقویت این اثرات همراه بوده است.

Introduction: Transforming growth factorβ1 is a key molecule in diabetes-induced renal fibrosis, and understanding the mechanisms of various interventions that protect renal tissue is importance. In this study, changes in Transforming growth factor β1 and Sirtuin1 gene expression in kidney tissue following high-intensity interval training(HIIT) and Fisetin were investigated in type 2 diabetic (T2D) mice.
Materials and Methods: 30 male C57bl/6 mice were divided into control groups with normal diet (n=6) and high-fat diet (n=24). T2D was induced by 16 weeks of high-fat diet. Then, diabetic mice were randomly divided into diabetes, diabetes-exercise, diabetes-Fisetin, and diabetes-exercise-Fisetin groups. Fisetin was injected intraperitoneally daily at a dose of 20 mg/kg for 8 weeks.
The HIIT protocol consisted of 8 weeks of running with 2-minute intervals at an intensity of 80-110% of maximal aerobic power with 2-minute rest intervals at an intensity of 50% of maximal aerobic power, 5 days/week. Data analysis was performed using one-way analysis of variance (P<0.05).
Results: With the induction of diabetes, Transforming growth factor β1 gene expression in kidney tissue increased and Sirtuin1 decreased(P<0.001). Also, HIIT, Fisetin, and combined intervention led to significant changes in Transforming growth factor β1 and Sirtuin1 gene expression, but the combined intervention had a significantly stronger effect on these changes compared to the other two interventions (P<0.001).
Conclusion: It seems that HIIT and Fisetin can reduce kidney damage caused by T2D by increasing Sirtuin1, at least by reducing Transforming growth factor β1 gene expression. Although the combined intervention was associated with enhanced effects.
