چکیده
مقدمه : توکسوپلاسما گوندی عامل بیماری توکسوپلاسموزیس است و شکل اصلی بیماری زایی آن تاکی زوئیت می باشد. یکی از مهمترین سدهای دفاعی در مقابل این انگل ماکروفاژها هستند. ماکروفاژهای آلوده به توکسوپلاسما گوندی از طریق فعال شدن گیرنده P2X7R به حذف انگل کمک می کنند. هدف مطالعه حاضر بررسی بیان ژن P2X7R درسلولهای رده منوسیت/ماکروفاژی THP-1 مجاور شده با غلظت های مختلف آنتی ژن محلول تاکی زوئیت توکسوپلاسما گوندی بود.
مواد و روش ها: در مطالعه اخیر سلولهای رده ماکروفاژی THP-1 با غلظت های 100، 80، 40، 20 و 10 میکروگرم بر میلی لیتر آنتی ژن محلول تاکی زوئیت توکسوپلاسما گوندی به مدت چهار ساعت مجاور شدند. پس از استخراج RNA کلی و سنتز cDNA به روش Real time PCR بیان این ژن ارزیابی شد.
یافته ها: نتایج به دست آمده نشان داد که پس از چهار ساعت همه غلظت های آنتی ژن محلول تاکی زوئیت توکسوپلاسما گوندی در کشت سلولی تیمار شده سبب کاهش بیان ژن P2X7R می شوند. بیشترین کاهش بیان این ژن در غلظت 80 میکروگرم در میلی لیتر مشاهده شد. همچنین کاهش بیان ژن مذکور با غلظت آنتی ژن درگروه ها ارتباط معنا داری نداشت.
نتیجه گیری: نتایج این مطالعه نشان داد که آنتی ژن محلول تاکی زوئیت توکسوپلاسما گوندی سویه RH می تواند سبب کاهش بیان ژن P2X7R شود و احتمالا کمک به فرار انگل از سیستم ایمنی شود.

چکیده [English]
Introduction: Toxoplasma gondii is the causative agent of toxoplasmosis. Macrophages are one of the most important immune cells against toxoplasmosis. Activation of P2X7R plays a critical role during immune response through macrophages.This study aimed to evaluate the expression of P2X7R gene in THP-1 cell line treated by T. gondii soluble total antigen (STAg).
Materials and Methods: STAg was extracted from T. gondii tachyzoites strain RH. Upon transforming THP-1 cell line to macrophage M0, the cells were treated by different concentrations of STAg including 100, 80, 40, 20, and 10 µg for 4 h. Total RNA was extracted from cells, cDNA was synthesized, and relative quantitative real-time PCR was performed to evaluate the expression of P2X7R gene.
Results: Results of real-time PCR revealed that except concentrations 10 and 40 µg, the other concentrations of T. gondii STAg significantly decreased the expression of P2X7R gene. The highest downregulation of P2X7R gene was observed in THP-1 sensed by the concentration 80 µg.
Conclusion: In addition, there was no statistically correlation between the P2X7R gene expression and the concentration of T. gondii STAg. This study showed that T. gondii STAg my affect the expression of P2X7R to help the parasite to escape from immune responses leading by macrophages.
