تأثیر عصاره ریز جلبک اسپیرولینا بر روی بیان ژن ضد آپوپتوتیک Bcl-2 در رده سلولی سرطان مغز

چکیده

مقدمه: رده سلولی U87 شامل سلول ­های سرطانی گلیوبلاستوما به عنوان حادترین نوع تومور مغزی از رده انسانی می ­باشد. افزایش بیان ژن Bcl-2 در سلول ­های سرطانی اثر ضد آپوپتوزی دارد. در مطالعه حاضر اثر عصاره جلبک اسپیرولینا بر روی میزان بیان ژن در رده سلول سرطانی بررسی شده است.
روش کار: در این مطالعه آزمایشگاهی، از رده سلولی U87 استفاده گردید. در این مطالعه رده سلولی سرطان مغز از مرکز ذخایر زیستی و ژنتیکی ایران تهیه و کشت داده شد. سلول ­ها به دو گروه تیمار و شاهد تقسیم شدند. تأثیر عصاره جلبک اسپیرولینا روی گروه تیمار با استفاده از MTT  ( (compound
3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromidefor) )  سنجیده شد. ژن مرجع در این مطالعه GAPDH بود. از سلول ­های گروه تیمار و کنترل RNA استخراج و سپس cDNA ساخته شد. میزان نسبی بیان ژن­ های Bcl2 و GAPDH با استفاده ازReal-time PCR  و Ct∆∆ مورد بررسی قرار گرفت. نتایج با استفاده از نرم ­افزار REST 2009 نسخه 2013  تجزیه و تحلیل گردید.
یافته­ ها: نتیجه MTT  نشان داد که IC50 ( Median Inhibition Concentration) در 48 ساعت تیمار،  mg/ml 10.01  می ­باشد. با انجام Real-time PCR  مشخص شد که در این سلول­ ها، میزان بیان ژن ضد آپوپتوزی Bcl-2 به میزان 3 برابر کاهش معنی داری پیدا کرد.
نتیجه­ گیری: با توجه به نتایج می­ توان گفت که در سلول ­های گروه تیمار با عصاره جلبک میزان بیان Bcl-2 سه برابر و به­ طور معنی ­داری کاهش داشته است.

Introduction: U87 cell line contains glioblastoma cancer cells as the most acute type of human brain tumor. Increased
expression of Bcl-2 gene in cancer cells has anti-apoptotic effect. In the present study the effect of Spirulina extract on gene expression in cancer cell line was investigated.
Materials and Methods: In this in laboratory study, the U87 cell line was used. The brain cancer cell line was procured and
cultivated from the Iranian Biological Resource Center. Cells were divided into two treatment and control groups. Spirulina alga extract was affected on the treatment group and its toxicity was measured using MTT assay (compound 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromidefor). The reference gene in this study was GAPDH. RNA was extracted from control and control cells and cDNA was made. The expression of genes was evaluated using ∆∆Ct in Real-time PCR.
Results: The result of MTT assay showed that the IC50 (Median Inhibition Concentration) value was 10.01 mg/ml for the 48 hour treatment. The result of Real-time PCR showed that the expression of Bcl-2 and the anti-apoptotic gene significantly decreased 3 folds in treatment group.
Conclusion: According to the results of MTT assay and Real-time PCR, the expression of Bcl-2 significantly decreased 3 times in treatment group
