تأثیر دوازده هفته تمرین هوازی همراه با مکمل‌ یاری عصاره‌ی گل‌سرخ بر بیان ژن‌های کاسپاز 9 و کاسپاز3 عضله‌ی نعلی موش‌های صحرایی نر

چکیده

مقدمه: کاسپازها در فرایند مرگ برنامه‌ریزی شده سلول نقش مهمی دارند و از طرفی گل‌ سرخ نیز دارای تاثیرات محافظتی در این زمینه می‌باشد. هدف از مطالعه حاضر تعیین تاثیر تمرین ­های هوازی همراه با عصاره‌ی گل سرخ بر بیان ژن‌های کاسپاز 3 و 9 عضله‌ی نعلی در موش‌های صحرایی نر بود.
روش کار: در این تحقیق تجربی، 50 سر موش صحرایی نر دو ماهه با نژاد ویستار تهیه و به صورت تصادفی در پنج گروه (کنترل سه ماهه، کنترل شش ماهه، تمرین، عصاره گل سرخ، ترکیبی) قرار گرفتند. گروه‌های تمرین هوازی و تمرین هوازی+ گل سرخ به مدت 5 روز در هفته و به مدت 12 هفته روی نوارگردان به شیب 15 درصد شرکت کردند. گروه عصاره گل سرخ و ترکیبی از طریق گاواژ، به ازای هر کیلوگرم وزن بدن یک گرم (0.09 گرم گل سرخ + 9 سی‌سی سالین) دریافت نمودند. پس از مداخله تمرینات هوازی و مصرف عصاره گل سرخ، مراحل جراحی و استخراج نمونه برای موش‌ها انجام شد. داده‌های حاصل با استفاده از آزمون‌ آنالیز واریانس در سطح معنی ­داری کمتر از 0.50 تجزیه و تحلیل شدند.
یافته‌ها: بیان ژن کاسپاز 3 و 9 در عضله نعلی گروه­ های تمرین هوازی و عصاره گل سرخ و گروه ترکیبی (گل سرخ و تمرین هوازی) نسبت به گروه کنترل شش ماهه و سه ماهه بیان کمتری داشته است به طوری که در گروه­ کنترل شش ماهه بیشترین بیان را داشته است (0.05>P).
نتیجه‌گیری: با توجه به نتایج، احتمالا با استفاده از تمرینات هوازی و عصاره‌ی گل سرخ می ­توان میزان آپوپتوز سلولی را با کاهش بیان ژن کاسپاز 3 و 9 کاهش داد و روند ضعف عضلات بدن را نیز کند نمود.

Introduction: Caspases play an important role in the cellular death process and, on the other hand, Rose has protective effects in this area. The purpose of this study was to investigate the effect of aerobic exercises with Rose damascena supplementation on the expression of caspase 3 and 9 genes of soleus muscle in male rats.
Materials and Methods: In this experimental study, 50 male Wistar, two-month-old rats were randomly divided into five
groups (three-month control, six-month control, exercise, Rose damascena, and combined). Aerobic exercise and aerobic exercises + Rose participated in the treadmill on a gradient of 15% for 5 days a week and for 12 weeks, and the Rose damascena and combined groups received 0.09 grams of rose + 9 cc saline per kg body weight with gavage,. After intervention aerobic exercises and use of Rose damascena, surgical procedures and extraction of specimens were performed in rats. The data were analyzed by ANOVA at a significant level of less than 0.05.
Results: Caspase 3 and 9 genes expression in soleus muscles of aerobic exercise and rose damascena and combaind group (rose and aerobic exercise) was less pronounced than in six-month control group, respectively, so that in 6 month control group the highest expression was observed.
Conclusion: According to the results, aerobic exercises and Rose damascena maybe can reduce the rate of cell
apoptosis by reducing the expression of Caspase 3 and 9 genes, and slow down the muscle weakness.
